Our scientists have worked in the field of structural biology and x-ray crystallography including membrane protein crystallography and high-throughput methods. We have over 100 entries in the PDB and have purified over 100 unique proteins.
We provide both gene-to-structure and high-throughput co-crystallization services.
Working with our clients, we routinely assess all programs for feasibility and provide a research plan in advance of initiating work. We keep your eventual goal in mind and will work to provide the best system for your studies. For example, for some projects with known crystal structures, we have re-engineered proteins to obtain better diffracting crystals or to produce co-crystals with ligands where the native protein was not adequate. We also routinely work with proteins of unknown structure using a high-throughput cloning and expression system using multiple domain boundaries, vectors and expression systems.
Protein Production
Our molecular biology laboratory is fully equipped for expression in E coli and baculovirus. We have customized proprietary vectors in addition to commercially available to provide the best system for protein production. We produce protein for our own internal programs and structural biology contracts.
We will produce protein and provide to clients as a service.
Protein Crystallization
We have crystallized more than 15 new proteins for our clients since Zenobia was founded in mid-2008. Most diffract to < 2.5 angstrom resolution, and many diffract to ultra-high resolution 1.0 - 1.3 angstrom. We routinely screen over 1000 conditions in crystallization for each construct we prepare. An example of the map for an ultra-high resolution structure solved for one of our clients serves as the background image for our web-pages (see above).
We will conduct crystallization trials with client protein on a case by case basis, although we prefer to prepare the protein fresh at Zenobia.
Data collection and structure solution
We have routine access to third-generation
synchrotrons year-round which provides for rapid high-quality data for the crystals we obtain. Automated structure determination programs allow rapid processing of these data and conversion to high resolution x-ray structures. Power point reports and data statistics are routinely provided for each structure completed.
